{"id":12084,"date":"2015-12-26T10:29:38","date_gmt":"2015-12-26T10:29:38","guid":{"rendered":"https:\/\/genomecanada.ca\/drupal\/news_release\/percee-majeure-pour-developper-de-nouveaux-medicaments-anticancereux-pieger-les-cellules\/"},"modified":"2015-12-26T10:29:38","modified_gmt":"2015-12-26T10:29:38","slug":"percee-majeure-pour-developper-de-nouveaux-medicaments-anticancereux-pieger-les-cellules","status":"publish","type":"post","link":"https:\/\/genomecanada.ca\/fr\/percee-majeure-pour-developper-de-nouveaux-medicaments-anticancereux-pieger-les-cellules\/","title":{"rendered":"Perc\u00e9e majeure pour d\u00e9velopper de nouveaux m\u00e9dicaments anticanc\u00e9reux: Pi\u00e9ger les cellules souches leuc\u00e9miques"},"content":{"rendered":"<p>L\u2019Institut de recherche en immunologie et en canc\u00e9rologie (IRIC) de l\u2019Universit\u00e9 de Montr\u00e9al (UdeM) en collaboration avec la Banque de cellules leuc\u00e9miques du Qu\u00e9bec de l\u2019H\u00f4pital Maisonneuve-Rosemont a r\u00e9cemment r\u00e9alis\u00e9 une perc\u00e9e majeure gr\u00e2ce \u00e0 la culture de cellules souches leuc\u00e9miques en laboratoire, ce qui permettra d\u2019acc\u00e9l\u00e9rer le d\u00e9veloppement de nouveaux m\u00e9dicaments anticanc\u00e9reux.<\/p>\n<p>Dans une r\u00e9cente \u00e9tude publi\u00e9e dans Nature Methods, les scientifiques d\u00e9crivent comment ils ont r\u00e9ussi \u00e0 identifier deux nouveaux compos\u00e9s chimiques qui permettent le maintien des cellules souches leuc\u00e9miques lorsque celles-ci sont cultiv\u00e9es \u00e0 l\u2019ext\u00e9rieur du corps.<\/p>\n<p>Cette importante avanc\u00e9e ouvre la voie \u00e0 l\u2019identification de nouveaux m\u00e9dicaments anticanc\u00e9reux pour contrer la leuc\u00e9mie my\u00e9lo\u00efde aigu\u00eb, une des formes les plus agressives de cancer du sang.<\/p>\n<p>La capacit\u00e9 \u00e0 cultiver des cellules souches leuc\u00e9miques en culture repr\u00e9sente une perc\u00e9e majeure. La prochaine \u00e9tape est d\u2019\u00e9tudier les m\u00e9canismes mol\u00e9culaires qui contr\u00f4lent le maintien et la prolif\u00e9ration des cellules leuc\u00e9miques ainsi que la r\u00e9sistance aux m\u00e9dicaments anticanc\u00e9reux.<\/p>\n<p>Cette \u00e9tude est issue du Groupe de recherche Leuc\u00e9g\u00e8ne. Ce groupe est codirig\u00e9 par le Dr&nbsp;Guy Sauvageau, chef de la direction et chercheur principal \u00e0 l\u2019IRIC ainsi que professeur au D\u00e9partement de m\u00e9decine de l\u2019UdeM, la Dre Jos\u00e9e&nbsp;H\u00e9bert, directrice de&nbsp;la Banque&nbsp;de cellules leuc\u00e9miques du Qu\u00e9bec, h\u00e9matologue \u00e0 l\u2019H\u00f4pital Maisonneuve-Rosemont et professeure au D\u00e9partement de m\u00e9decine de l\u2019UdeM, et&nbsp;S\u00e9bastien Lemieux, chercheur principal \u00e0 l\u2019IRIC. La premi\u00e8re auteure de l\u2019\u00e9tude est Caroline Pabst, stagiaire postdoctorale \u00e0 l\u2019IRIC et associ\u00e9e au Groupe de recherche Leuc\u00e9g\u00e8ne.<\/p>\n<p>\u00ab&nbsp;Cette perc\u00e9e scientifique d\u00e9montre l\u2019avantage de travailler au sein d\u2019une \u00e9quipe multidisciplinaire comme celle du Groupe de recherche Leuc\u00e9g\u00e8ne. L\u2019acc\u00e8s \u00e0 des cellules de patients atteints de leuc\u00e9mie et \u00e0 des installations \u00e0 la fine pointe de la technologie de l\u2019IRIC sont \u00e9galement des atouts essentiels pour mener \u00e0 bien des recherches innovatrices&nbsp;\u00bb, ajoutent les Drs Guy Sauvageau et Jos\u00e9e H\u00e9bert.<\/p>\n<p>Mise en contexte de l\u2019\u00e9tude<\/p>\n<p>Les cellules souches situ\u00e9es dans la moelle osseuse servent \u00e0 la production des cellules sanguines. Malheureusement, la d\u00e9r\u00e9gulation de ces cellules a souvent des cons\u00e9quences d\u00e9sastreuses lorsque l\u2019une d\u2019elles d\u00e9veloppe des mutations qui la transforment en une cellule souche maligne dite \u00ab&nbsp;leuc\u00e9mique&nbsp;\u00bb.&nbsp;<br \/>\n&nbsp;<\/p>\n<p>Il en r\u00e9sulte une prolif\u00e9ration anormale des cellules du sang et le d\u00e9veloppement d\u2019une leuc\u00e9mie. Les cellules souches leuc\u00e9miques sont \u00e9galement une des causes probables de rechute des patients, car elles sont particuli\u00e8rement r\u00e9sistantes aux traitements anticanc\u00e9reux.<\/p>\n<p>L\u2019obstacle majeur avant cette d\u00e9couverte \u00e9tait de cultiver des cellules souches et de les maintenir&nbsp;in vitro, car elles perdaient rapidement leur caract\u00e8re de cellules souches canc\u00e9reuses.
