D.<\/em><\/strong><\/h2>\n<p>Pour suivre l\u2019\u00e9volution du SRAS-CoV-2 et orienter la r\u00e9ponse du Canada \u00e0 la COVID-19, il a fallu accro\u00eetre la capacit\u00e9 de s\u00e9quen\u00e7age g\u00e9nomique et d\u2019\u00e9change des donn\u00e9es au pays. Nous avons demand\u00e9 \u00e0\u00a0<a href=\"https:\/\/www.ualberta.ca\/medicine\/about\/people\/details.html?n=matthew-croxen\" target=\"_blank\" rel=\"noopener noreferrer\">Matthew Croxen<\/a>, Ph.\u00a0D., copr\u00e9sident du Groupe de travail sur le renforcement des capacit\u00e9s du R\u00e9seau canadien de g\u00e9nomique COVID-19 (RCanG\u00e9CO), de nous parler de l\u2019\u00e9volution du s\u00e9quen\u00e7age au cours de la pand\u00e9mie et de ce qu\u2019elle signifie pour les Canadiennes et les Canadiens.<\/p>\n<p>M.\u00a0Croxen est responsable de la g\u00e9nomique en sant\u00e9 publique (Alberta Precision Laboratories) et professeur adjoint au D\u00e9partement de m\u00e9decine de laboratoire et de pathologie de la Facult\u00e9 de m\u00e9decine et de dentisterie de la University of Alberta.<\/p>\n<hr \/>\n<h3><em>\u00ab\u2009\u00c0 mon avis, il y a maintenant plus un changement de cap et une prise de conscience que la g\u00e9nomique est le prochain outil de la surveillance des agents pathog\u00e8nes pour les laboratoires de sant\u00e9 publique [\u2026] La capacit\u00e9 que nous avons mise en place gr\u00e2ce au RCanG\u00e9CO, et en g\u00e9n\u00e9ral tout au long de la pand\u00e9mie, peut servir pour d\u2019autres maladies infectieuses telles que les pathog\u00e8nes d\u2019origine alimentaire, l\u2019influenza, la tuberculose et les bact\u00e9ries r\u00e9sistantes aux antimicrobiens.\u2009\u00bb<\/em><\/h3>\n<p><em>\u2013 Matthew Croxen, Ph. D.<\/em><\/p>\n<hr \/>\n<h2><strong>D\u00e9crivez-nous votre travail dans le RCanG\u00e9CO?<\/strong><\/h2>\n<p>Au cours des deux derni\u00e8res ann\u00e9es, nous avons particip\u00e9 au renforcement de la capacit\u00e9 de s\u00e9quen\u00e7age g\u00e9nomique \u2014 depuis l\u2019octroi de financement pour le mat\u00e9riel de s\u00e9quen\u00e7age jusqu\u2019\u00e0 la formation du personnel \u2014 ici en Alberta et partout ailleurs au Canada, pour r\u00e9ussir \u00e0 contrer les variants du SRAS-CoV-2 et les pand\u00e9mies futures. Au d\u00e9but de la pand\u00e9mie, le renforcement de la capacit\u00e9 de s\u00e9quen\u00e7age \u00e9tait le besoin le plus urgent dans les laboratoires de sant\u00e9 publique provinciaux et territoriaux du pays. Nous avons \u00e9galement reconnu qu\u2019il fallait aussi doter certains h\u00f4pitaux d\u2019une petite capacit\u00e9 de s\u00e9quen\u00e7age.<\/p>\n<h2><strong>Comment la capacit\u00e9 de s\u00e9quen\u00e7age de l\u2019Alberta a-t-elle chang\u00e9 au cours de la pand\u00e9mie?<\/strong><\/h2>\n<p>Nous avions un peu de capacit\u00e9 de s\u00e9quen\u00e7age au d\u00e9but de la pand\u00e9mie, mais pas beaucoup. Il y avait un technologue qui s\u2019effor\u00e7ait de comprendre la m\u00e9thodologie du laboratoire et de s\u2019habituer au s\u00e9quen\u00e7age d\u2019un nouveau virus \u00e0 ARN dont nous savions tr\u00e8s peu de choses. Nous n\u2019avions \u00e0 ce moment-l\u00e0 qu\u2019une capacit\u00e9 de s\u00e9quen\u00e7age \u00e0 Edmonton. Nous effectuons maintenant du s\u00e9quen\u00e7age dans des laboratoires dot\u00e9s de divers s\u00e9quenceurs \u00e0 Calgary et \u00e0 Edmonton. Nous disposons aussi aux deux endroits d\u2019un plus grand nombre de personnes form\u00e9es et nous s\u00e9quen\u00e7ons entre 800 et 1\u2009000\u00a0g\u00e9nomes de SRAS-CoV-2 par semaine, gr\u00e2ce \u00e0 cette capacit\u00e9.<\/p>\n<h2><strong>Comment le s\u00e9quen\u00e7age g\u00e9nomique a-t-il orient\u00e9 la r\u00e9ponse de la sant\u00e9 publique de l\u2019Alberta \u00e0 la COVID-19?<\/strong><\/h2>\n<p>Nous avons pu rapidement fournir de l\u2019information sur le virus et les nouveaux variants pr\u00e9occupants. Le s\u00e9quen\u00e7age nous donne un aper\u00e7u d\u00e9taill\u00e9 de la composition g\u00e9n\u00e9tique du virus, ce qui nous permet de rep\u00e9rer les diff\u00e9rences dans le profil mutationnel d\u2019un \u00e9chantillon donn\u00e9. Nous pouvons aussi d\u00e9terminer si un virus SRAS-CoV-2 est un variant pr\u00e9occupant, ce qui a vraiment contribu\u00e9 \u00e0 orienter la sant\u00e9 publique, car nous pouvons rapidement savoir quels variants circulent. Lorsque le variant Alpha (B. 1. 1. 7) a \u00e9t\u00e9 annonc\u00e9 la premi\u00e8re fois au Royaume-Uni, notre s\u00e9quen\u00e7age nous a permis de rechercher le profil mutationnel dans les g\u00e9nomes du SRAS-CoV-2 recueillis au Canada, confirmant ainsi que le variant Alpha circulait ici aussi.<\/p>\n<p>En Alberta, nous avons aussi associ\u00e9 le s\u00e9quen\u00e7age \u00e0 un test de d\u00e9pistage rapide des variants. Ce test est rapide et peut d\u00e9tecter diff\u00e9rents variants pr\u00e9occupants. Quand on a commenc\u00e9 \u00e0 parler d\u2019Omicron, Delta \u00e9tait encore le principal variant en circulation au Canada. Comme nous pouvions utiliser ce test de d\u00e9pistage rapide pour identifier les variants Delta, nous avons pu concentrer nos efforts de s\u00e9quen\u00e7age sur les autres variants que Delta, ce qui nous a aid\u00e9s \u00e0 reconna\u00eetre rapidement la pr\u00e9sence d\u2019Omicron en Alberta.<\/p>\n<h2><strong>Le Portail canadien de donn\u00e9es de VirusSeq a \u00e9t\u00e9\u00a0<\/strong><a href=\"https:\/\/www.genomecanada.ca\/fr\/nouvelles\/genome-canada-lance-le-portail-national-de-donnees-pour-suivre-la-covid-19-en-temps-reel\" target=\"_blank\" rel=\"noopener noreferrer\"><strong>lanc\u00e9 en avril\u00a02021<\/strong><\/a><strong>\u00a0pour faciliter l\u2019\u00e9change de donn\u00e9es du SRAS-CoV-2 partout au Canada et aider les chercheurs \u00e0 r\u00e9soudre des probl\u00e8mes futurs de sant\u00e9 publique. Comment les chercheurs utilisent-ils ce nouvel outil?<\/strong><\/h2>\n<p>L\u2019Alberta et la plupart des provinces contribuent au\u00a0<a href=\"https:\/\/virusseq-dataportal.ca\/\" target=\"_blank\" rel=\"noopener noreferrer\">Portail canadien de donn\u00e9es de VirusSeq<\/a>. Il s\u2019agit d\u2019une solution de rechange \u00e0 une autre archive publique appel\u00e9e\u00a0<a href=\"https:\/\/www.gisaid.org\/\" target=\"_blank\" rel=\"noopener noreferrer\">GISAID<\/a>, qui h\u00e9berge des millions de g\u00e9nomes du SRAS-CoV-2 de partout dans le monde. Le Portail canadien de donn\u00e9es VirusSeq n\u2019h\u00e9berge que des g\u00e9nomes canadiens du SRAS-CoV-2. Il s\u2019agit en quelque sorte d\u2019un guichet unique d\u2019acc\u00e8s aux donn\u00e9es de s\u00e9quen\u00e7age canadiennes, ce qui \u00e9vite d\u2019avoir \u00e0 trier tous les g\u00e9nomes du SRAS-CoV-2 du monde dans GISAID.<\/p>\n<p>Le portail de VirusSeq est public, de sorte que les chercheurs ont un acc\u00e8s libre aux donn\u00e9es pour leurs travaux. Des chercheurs de tout le Canada \u00e9tudient certaines des diff\u00e9rentes lign\u00e9es du SRAS-CoV-2 et tentent de mod\u00e9liser la valeur s\u00e9lective et les taux de croissance de certains variants dans diff\u00e9rentes provinces. Il y a quelques jours, un chercheur m\u2019a contact\u00e9 pour me poser une question sur la fr\u00e9quence d\u2019une certaine mutation dans son g\u00e8ne cible. J\u2019ai pu rapidement, en quelques heures, t\u00e9l\u00e9charger les 180\u2009000\u00a0g\u00e9nomes canadiens du portail de VirusSeq et lui communiquer la distribution des mutations dans ce g\u00e8ne \u00e0 partir des virus SRAS-CoV-2 circulant au Canada depuis le d\u00e9but de la pand\u00e9mie. VirusSeq est un outil puissant pour r\u00e9pondre aux questions des chercheurs, offrant un acc\u00e8s rapide et ouvert au contexte canadien de la pand\u00e9mie de SRAS-CoV-2.<\/p>\n<h2><strong>Les activit\u00e9s et les investissements du RCanG\u00e9CO ces deux derni\u00e8res ann\u00e9es auront-ils des r\u00e9percussions au-del\u00e0 de la pand\u00e9mie actuelle?<\/strong><\/h2>\n<p>\u00c0 mon avis, il y a maintenant plus un changement de cap et une prise de conscience que la g\u00e9nomique est le prochain outil de la surveillance des agents pathog\u00e8nes pour les laboratoires de sant\u00e9 publique. Le s\u00e9quen\u00e7age des g\u00e9nomes fournit beaucoup plus d\u2019information que les m\u00e9thodes traditionnelles de typage utilis\u00e9es pour la surveillance des agents pathog\u00e8nes, souvent limit\u00e9e \u00e0 une petite portion du g\u00e9nome et ne comportant souvent aucun s\u00e9quen\u00e7age. La capacit\u00e9 que nous avons mise en place gr\u00e2ce au RCanG\u00e9CO, et en g\u00e9n\u00e9ral tout au long de la pand\u00e9mie, peut servir pour d\u2019autres maladies infectieuses telles que les pathog\u00e8nes d\u2019origine alimentaire, l\u2019influenza, la tuberculose et les bact\u00e9ries r\u00e9sistantes aux antimicrobiens. C\u2019est l\u00e0 un aspect important du travail du Groupe de travail sur le renforcement des capacit\u00e9s du RCanG\u00e9CO\u00a0: augmenter la capacit\u00e9 pour le SRAS-CoV-2, mais jeter aussi les bases de la surveillance d\u2019autres agents pathog\u00e8nes. Un grand nombre des laboratoires de sant\u00e9 publique du Canada sont maintenant capables d\u2019accepter diff\u00e9rentes priorit\u00e9s de surveillance d\u2019autres maladies infectieuses importantes. Nous disposerons de plus d\u2019outils et de capacit\u00e9s pour mieux surveiller les maladies infectieuses, mais aussi travailler en collaboration, comme nous l\u2019avons fait pour le SRAS-CoV-2.<\/p>\n<p>Nous sommes fiers de ce que nous avons accompli pour relever le d\u00e9fi de la COVID-19, mais il y a encore beaucoup \u00e0 faire et de grandes questions encore sans r\u00e9ponse. Comment allons-nous maintenir cet \u00e9lan? Comment le rendrons-nous durable et continuerons-nous de travailler ensemble? Le financement du RCanG\u00e9CO pour le renforcement des capacit\u00e9s a aid\u00e9 \u00e0 jeter les bases de la recherche de solutions \u00e0 toutes ces questions et donn\u00e9 aux laboratoires canadiens de sant\u00e9 publique les moyens de r\u00e9ussir \u00e0 l\u2019avenir.<\/p>\n<hr \/>\n<div class=\"l-content\" role=\"main\">\n<div class=\"l-content-wrapper\">\n<article class=\"node node--blog node--full node--blog--full\" role=\"article\">\n<div class=\"node__content\">\n<div class=\"field field--name-body field--type-text-with-summary field--label-hidden\">\n<div class=\"field__items\">\n<div class=\"field__item even\">\n<p><em><a href=\"https:\/\/genomecanada.ca\/fr\/domaines-defis\/rcangeco\/\" rel=\"noopener noreferrer\">Le R\u00e9seau canadien de g\u00e9nomique COVID-19 (RCanG\u00e9CO)<\/a>\u00a0a pour mission de relever le d\u00e9fi de la COVID-19 en produisant les donn\u00e9es accessibles et utilisables des g\u00e9nomes viraux et humains pour orienter les d\u00e9cisions strat\u00e9giques et les d\u00e9cisions en sant\u00e9 publique, et mettre au point des traitements et des vaccins.
