Ce lien est tout particuli\u00e8rement important chez les humains pour lesquels les chercheurs s\u2019efforcent de d\u00e9finir le fondement g\u00e9n\u00e9tique des maladies les plus courantes. M\u00eame si leur objectif ultime est de conna\u00eetre ces liens chez les humains, les chercheurs approfondiront la majeure partie des d\u00e9tails chez les souris, un organisme plus facile \u00e0 manipuler g\u00e9n\u00e9tiquement. Le pourcentage des similitudes entre les s\u00e9quences des humains et celles des souris d\u00e9passe 99 %. On croit donc que la compr\u00e9hension du fondement g\u00e9n\u00e9tique des maladies chez la souris orientera les recherches chez les humains.<\/p>\n<p>Pour d\u00e9finir le lien entre les g\u00e8nes de la souris et leur fonction dans l\u2019organisme, un programme financ\u00e9 \u00e0 l\u2019\u00e9chelle internationale a pour objet de muter tous les g\u00e8nes de la souris (environ 25 000), un g\u00e8ne \u00e0 la fois. On obtiendra de cette mani\u00e8re 25 000 souris diff\u00e9rentes pour lesquelles il faudra se poser la question suivante : \u00ab Comment ces souris se distinguent-elles d\u2019une souris normale dont tous les g\u00e8nes sont intacts? \u00bb<\/p>\n<p>Les chercheurs du pr\u00e9sent projet d\u00e9velopperont une nouvelle technologie fond\u00e9e sur l\u2019imagerie et l\u2019analyse informatique automatis\u00e9e qui permettra de visualiser les embryons et de les comparer \u00e0 un embryon qui n\u2019aura pas fait l\u2019objet d\u2019une mutation. L\u2019imagerie tridimensionnelle fera appel \u00e0 l\u2019imagerie par r\u00e9sonance magn\u00e9tique (IRM) qui joue un r\u00f4le important dans les diagnostics chez les humains en raison de la capacit\u00e9 de voir des tissus mous. Les chercheurs examineront \u00e9galement les possibilit\u00e9s d\u2019un autre type d\u2019imagerie optique tridimensionnelle parce que cette derni\u00e8re peut avoir une meilleure r\u00e9solution et une sensibilit\u00e9 exceptionnelle aux tissus. Pour ces deux types d\u2019imagerie, les chercheurs \u00e9laboreront une analyse informatique perfectionn\u00e9e pour comparer les images d\u2019un embryon dans lequel une seule mutation a \u00e9t\u00e9 faite avec un embryon t\u00e9moin et ainsi observer les r\u00e9gions o\u00f9 apparaissent des diff\u00e9rences. Cette technologie est essentielle pour que les 25 000 animaux g\u00e9n\u00e9tiquement modifi\u00e9s soient utiles. Dans deux ans, nous aurons d\u00e9termin\u00e9 le meilleur moyen de prendre des images des embryons pour qu\u2019un ordinateur puisse trouver la plupart des diff\u00e9rences. Cette technologie sera ensuite mise \u00e0 la disposition des centres de g\u00e9nomique du Canada et deviendra un service offert dans des installations de g\u00e9nomique dans d\u2019autres pays.
