Un segment de 648 paires de bases du cytochrome c oxydase 1 (CO1) mitochondrial a maintenant \u00e9t\u00e9 choisi comme r\u00e9gion de base du code \u00e0 barres des eucaryotes. L\u2019examen horizontal de la diversit\u00e9 des s\u00e9quences dans cette r\u00e9gion g\u00e9n\u00e9tique est important dans de nombreux contextes : il permet d\u2019identifier et de d\u00e9couvrir des esp\u00e8ces; il r\u00e9v\u00e8le des facteurs qui influencent les rythmes de l\u2019\u00e9volution mol\u00e9culaire et l\u2019\u00e2ge des esp\u00e8ces; et il permet d\u2019\u00e9tudier en d\u00e9tail les voies \u00e9volutives de la prot\u00e9ine CO1. Pour ces raisons, on planifie actuellement un important programme international de recherche qui vise \u00e0 \u00e9toffer rapidement la biblioth\u00e8que de r\u00e9f\u00e9rence des s\u00e9quences de CO1. Le projet international du codage \u00e0 barres du vivant (iBOL), un programme quinquennal de 150 millions de dollars dirig\u00e9 par le Canada, fera intervenir des chercheurs de 25 pays. Pendant cette p\u00e9riode, iBOL livrera les codes \u00e0 barres de 500 000 esp\u00e8ces et les efforts subs\u00e9quents serviront \u00e0 produire une biblioth\u00e8que de r\u00e9f\u00e9rence des codes \u00e0 barres de tous les eucaryotes.<\/p>\n<p>M\u00eame s\u2019il peut falloir 20 ans pour constituer une biblioth\u00e8que de tous les eucaryotes, le codage \u00e0 barres de l\u2019ADN trouve d\u00e9j\u00e0 des applications, car des biblioth\u00e8ques de codes \u00e0 barres seront bient\u00f4t termin\u00e9es dans diff\u00e9rents groupes. On sait maintenant que l\u2019identification d\u2019esp\u00e8ces constitue un domaine d\u2019application particuli\u00e8rement important du codage \u00e0 barres de l\u2019ADN. Il est \u00e9galement \u00e9vident que ces applications feront intervenir deux courants technologiques diff\u00e9rents : analyse au point de contact de sp\u00e9cimens uniques et criblage de masse de codes \u00e0 barres. Il faudra des appareils d\u2019analyse imm\u00e9diate \u00e0 utiliser au point de contact pour identifier des esp\u00e8ces dans divers contextes comme les inspections dans les ports et la lutte contre les insectes ravageurs.<\/p>\n<p>Par contraste, le deuxi\u00e8me volet technologique, au c\u0153ur du pr\u00e9sent projet, permettra d\u2019analyser des \u00e9chantillons biotiques mixtes, mais moins rapidement. Nous soulignons que de nombreux eucaryotes sont trop petits, trop nombreux ou trop rapproch\u00e9s pour qu\u2019on puisse les analyser au moyen des protocoles traditionnels du codage \u00e0 barres. Nous surmonterons cet obstacle en \u00e9laborant les protocoles n\u00e9cessaires \u00e0 l\u2019analyse de n\u2019importe quelle collection d\u2019eucaryotes. Nous appelons cette approche \u00ab codage \u00e0 barres d\u2019\u00e9chantillons environnementaux \u00bb et nous sommes convaincus que sa mise en \u0153uvre r\u00e9side dans le couplage de technologies de s\u00e9quen\u00e7age parall\u00e8le en masse et de nouveaux outils informatiques. Ces analyses feront certainement progresser la surveillance de la biodiversit\u00e9.<\/p>\n<p>Il suffit d\u2019imaginer la nouvelle capacit\u00e9 perfectionn\u00e9e de surveiller la qualit\u00e9 de l\u2019environnement qui devient possible si nous pouvions rapidement obtenir de l\u2019information sur la composition des esp\u00e8ces de tout \u00e9chantillon environnemental. Les r\u00e9percussions de nos travaux sont toutefois plus larges \u2013 le codage \u00e0 barres d\u2019\u00e9chantillons environnementaux repr\u00e9sente l\u2019outil m\u00e9tag\u00e9nomique de la vie eucaryote. Nous recherchons du financement pour mettre au point le codage \u00e0 barres d\u2019\u00e9chantillons environnementaux qui constituera un volet technologique que les plateformes de s\u00e9quen\u00e7age pourront adopter. Nous utiliserons les nouveaux s\u00e9quenceurs en parall\u00e8le, mais il faudra d\u2019importantes innovations technologiques pour qu\u2019ils puissent prendre en charge le codage \u00e0 barres d\u2019\u00e9chantillons environnementaux. Il faudra en particulier de nouveaux outils informatiques pour analyser les donn\u00e9es des s\u00e9quences et de nouveaux protocoles pour r\u00e9cup\u00e9rer les codes \u00e0 barres de grands \u00e9chantillons mixtes. Nous ne pr\u00e9voyons pas venir \u00e0 bout de ce d\u00e9fi, mais nous pr\u00e9voyons que nos travaux d\u00e9montreront comment le codage \u00e0 barres d\u2019\u00e9chantillons environnementaux peut r\u00e9pondre \u00e0 un besoin r\u00e9el, \u00e0 savoir la biosurveillance des eaux int\u00e9rieures du Canada.<\/p>\n<p>Nous effectuerons ces travaux en \u00e9troite collaboration avec des chercheurs d\u2019Environnement Canada qui non seulement mettront \u00e0 profit leur exp\u00e9rience approfondie des \u00e9chantillons environnementaux, mais seront \u00e9galement d\u2019importants utilisateurs ultimes de la technologie. Nous collaborerons \u00e9galement avec des coll\u00e8gues du Genome Technology Centre de Stanford qui sont des chefs de file mondiaux dans les technologies de s\u00e9quen\u00e7age parall\u00e8le de masse.
