Les g\u00e8nes, les \u00ab unit\u00e9s d\u2019information \u00bb que contient le g\u00e9nome, sont des instructions pour produire des prot\u00e9ines, les outils de travail mol\u00e9culaires des syst\u00e8mes biologiques. Les progr\u00e8s r\u00e9cents en prot\u00e9omique (\u00e9tude des prot\u00e9ines) ont permis de d\u00e9couvrir et d&rsquo;analyser de grands nombres de prot\u00e9ines individuelles. Cependant, \u00e0 de nombreux \u00e9gards, la prot\u00e9omique soul\u00e8ve des d\u00e9fis technologiques et analytiques plus difficiles \u00e0 relever que la g\u00e9nomique, \u00e0 la fois en raison des grands volumes (de 20 000 \u00e0 25 000 g\u00e8nes comparativement \u00e0 environ un million de prot\u00e9ines chez les humains) et de la fragilit\u00e9 beaucoup plus grande des mol\u00e9cules des prot\u00e9ines que de l\u2019ADN. Il s\u2019est av\u00e9r\u00e9 tr\u00e8s difficile de traiter en parall\u00e8le un grand nombre de prot\u00e9ines en vue d\u2019applications m\u00e9dicales en raison des possibilit\u00e9s combinatoires qui d\u00e9coulent des affinit\u00e9s crois\u00e9es dans un grand nombre de mol\u00e9cules candidates et leur propension \u00e0 la liaison non sp\u00e9cifique. Voil\u00e0 pourquoi les progr\u00e8s technologiques en prot\u00e9omique se sont faits \u00e0 un rythme assez lent (comparativement \u00e0 la g\u00e9nomique) et que des progr\u00e8s importants dans les soins de sant\u00e9 tardent \u00e9galement \u00e0 se produire.<\/p>\n<p>Malgr\u00e9 de nombreuses tentatives de groupes partout dans le monde, la r\u00e9ussite jusqu\u2019\u00e0 maintenant dans ce domaine a \u00e9t\u00e9 au mieux modeste, en raison de la difficult\u00e9 du d\u00e9fi \u00e0 relever. Le pr\u00e9sent projet r\u00e9unit une \u00e9quipe interdisciplinaire tr\u00e8s solide qui propose une solution technologique tr\u00e8s novatrice \u00e0 ce d\u00e9fi. L\u2019\u00e9quipe se fonde sur les travaux \u00e9prouv\u00e9s r\u00e9cents de ses membres.<\/p>\n<p>Il est propos\u00e9, dans le pr\u00e9sent projet, de d\u00e9velopper une technologie pour d\u00e9pister avec efficacit\u00e9 et fiabilit\u00e9 un grand nombre de candidats pour l\u2019identification de biomarqueurs prot\u00e9iques de chacune des maladies et de miniaturiser la technologie pour en permettre un usage r\u00e9pandu. Le projet comprend une approche en deux volets pour relever ce d\u00e9fi difficile. Premi\u00e8rement, en se basant sur la technologie r\u00e9cemment mise au point \u00e0 McGill et \u00e0 l\u2019Universit\u00e9 de Sherbrooke pour cibler de grands nombres de prot\u00e9ines candidates bas\u00e9s sur les r\u00e9seaux de colocalisation et la d\u00e9tection multimodale, l\u2019\u00e9quipe \u00e9laborera une plateforme capable d\u2019analyser en parall\u00e8le de grands volumes de mol\u00e9cules candidates \u2013 avec une tr\u00e8s haute sp\u00e9cificit\u00e9 et sensibilit\u00e9. Cette plateforme sera au service des chercheurs de G\u00e9nome Qu\u00e9bec et de G\u00e9nome Canada. Deuxi\u00e8mement, en se basant sur la technologie r\u00e9cemment mise au point \u00e0 McGill et \u00e0 l\u2019IMI, une version miniaturis\u00e9e tr\u00e8s facile \u00e0 transporter de la technologie bas\u00e9e sur des microfluides num\u00e9riques et des instruments optiques int\u00e9gr\u00e9s sera mise \u00e0 la disposition de groupes de recherche partenaires pour l\u2019analyse sur place de prot\u00e9ines candidates dans des applications cibl\u00e9es.<\/p>\n<p>\u00c0 court terme, la plateforme influencera la d\u00e9couverte de biomarqueurs, car elle comblera l\u2019\u00e9cart entre la d\u00e9couverte de biomarqueurs par spectrom\u00e9trie de masse et les diagnostics cliniques. La plateforme vis\u00e9e, qui permettra d\u2019analyser en parall\u00e8le 100 segments, pourrait r\u00e9volutionner les biomarqueurs et leur validation parce qu\u2019elle pourra facilement \u00eatre utilis\u00e9e en laboratoire. Cette plateforme prot\u00e9omique int\u00e9gr\u00e9e est tr\u00e8s largement ax\u00e9e sur la technologie, plus pr\u00e9cis\u00e9ment sur l\u2019\u00e9laboration et l\u2019int\u00e9gration de plateformes habilitantes. Au cours de ce projet, nous pr\u00e9voyons qu\u2019il n\u2019y aura pas de grands enjeux en GE<sup>3<\/sup> DS (enjeux de la recherche en g\u00e9nomique li\u00e9s \u00e0 l\u2019\u00e9thique, \u00e0 l\u2019environnement, \u00e0 l\u2019\u00e9conomie, au droit et \u00e0 la soci\u00e9t\u00e9) dont il faudrait tenir compte et ne diff\u00e9reront pas de ceux que font na\u00eetre les pratiques de recherche habituelles en sciences biom\u00e9dicales.
