L\u2019\u00e9chantillon d\u2019un patient contient de nombreuses cellules diff\u00e9rentes, dont chacune peut \u00eatre distingu\u00e9e et identifi\u00e9e par des biomarqueurs (prot\u00e9ines, g\u00e8nes et petites mol\u00e9cules) qui constituent la signature biochimique d\u2019une cellule.<\/p>\n<p>Des recherches novatrices en leuc\u00e9mie ont permis de faire l\u2019hypoth\u00e8se qu\u2019une maladie donn\u00e9e peut \u00eatre aliment\u00e9e par une petite fraction seulement de cellules malades (\u00ab cellules souches canc\u00e9reuses \u00bb dans le cas du cancer), et qu\u2019il est par cons\u00e9quent particuli\u00e8rement difficile de les d\u00e9tecter et de les traiter. La signature des biomarqueurs refl\u00e8te \u00e9galement les processus biochimiques intercellulaires qui d\u00e9terminent l\u2019activit\u00e9 et le sort de ces derni\u00e8res. Par cons\u00e9quent, la capacit\u00e9 de mesurer l\u2019enti\u00e8re signature des biomarqueurs en m\u00eame temps pour des cellules individuelles permet de mieux comprendre la gen\u00e8se des cellules et de mettre au point des m\u00e9dicaments qui traiteront la maladie<\/p>\n<p>On fait d&rsquo;\u00e9normes progr\u00e8s relativement \u00e0 l\u2019identification des signatures des biomarqueurs pour les diagnostics et la compr\u00e9hension des interactions des biomarqueurs. Malheureusement, il y a peu d\u2019outils d\u2019analyse capables de reconna\u00eetre ces signatures, et ces outils ne parviennent pas \u00e0 d\u00e9tecter de nombreux biomarqueurs en une seule analyse. Les technologies actuelles de diagnostic pour les cellules isol\u00e9es sont fond\u00e9es sur la d\u00e9tection de la fluorescence de r\u00e9actifs \u00e9tiquet\u00e9s qui reconnaissent sp\u00e9cifiquement les biomarqueurs. M\u00eame s\u2019il est possible de surveiller jusqu\u2019\u00e0 dix voies de d\u00e9tection en m\u00eame temps, l\u2019approche est limit\u00e9e par la faible r\u00e9solution, ce qui entra\u00eene un chevauchement des signaux et d\u2019importantes erreurs lorsque les biomarqueurs sont pr\u00e9sents dans une large gamme de concentrations.<\/p>\n<p>Une nouvelle technologie s\u2019impose pour d\u00e9terminer simultan\u00e9ment du point de vue quantitatif et ind\u00e9pendamment jusqu\u2019\u00e0 100 biomarqueurs dans des cellules individuelles, en particulier lorsque cette analyse peut se faire \u00e0 haute vitesse, de fa\u00e7on \u00e0 analyser 1 000 cellules ou plus \u00e0 la seconde. Les candidats \u00e9laborent une solution novatrice \u00e0 cette difficult\u00e9 qui suscite beaucoup d\u2019enthousiasme dans la communaut\u00e9 scientifique. Leur approche consiste \u00e0 tirer profit de la haute r\u00e9solution de la spectrom\u00e9trie de masse pour faire la distinction entre des atomes de m\u00e9taux rares qui remplacent les colorants fluorescents actuellement utilis\u00e9s en biologie. Une nouvelle g\u00e9n\u00e9ration de r\u00e9actifs diagnostiques qui lient diff\u00e9rents m\u00e9taux aux biomarqueurs est en cours d\u2019\u00e9laboration. Ces\u00ab m\u00e9taux d\u2019\u00e9tiquetage \u00bb sont d\u00e9tect\u00e9s en nombre par un prototype de cytom\u00e8tre en flux de haute sensibilit\u00e9 et de haute r\u00e9solution. Cet instrument introduit des cellules individuelles \u00e0 un rythme rapide, jusqu\u2019\u00e0 1 000 cellules \u00e0 la seconde, dans un spectrom\u00e8tre de masse \u00e0 voies multiples. Dans notre projet actuel de g\u00e9nomique appliqu\u00e9e \u00e0 la sant\u00e9 humaine financ\u00e9 par G\u00e9nome Canada, nous avons pu d\u00e9montrer la faisabilit\u00e9 de cette nouvelle technologie.<\/p>\n<p>La pr\u00e9sente proposition vise \u00e0 transformer ce prototype d\u2019instrument de recherche complexe (ainsi que les r\u00e9actifs n\u00e9cessaires \u00e0 son fonctionnement) en un prototype technique qui sera mis \u00e0 la disposition d\u2019autres chercheurs de G\u00e9nome Canada et qui sera ensuite converti en un instrument commercial qui pourra servir au diagnostic et \u00e0 la recherche en g\u00e9n\u00e9ral.<\/p>\n<p>En plus de permettre aux chercheurs en g\u00e9nomique et en prot\u00e9omique d\u2019am\u00e9liorer consid\u00e9rablement la profondeur et l\u2019\u00e9tendue de l\u2019analyse cellulaire, le pr\u00e9sent projet fournira un outil diagnostique qui d\u00e9finira une nouvelle norme en mati\u00e8re de soins dans les h\u00f4pitaux, les cliniques et les services de recherche partout dans le monde.<\/p>\n<p>Les candidats poss\u00e8dent une exp\u00e9rience approfondie de l\u2019\u00e9laboration d\u2019outils d\u2019analyse qui font leurs preuves sur le plan commercial et sont dans une position id\u00e9ale pour r\u00e9aliser les objectifs ambitieux de ce projet. La r\u00e9ussite du projet m\u00e8nera \u00e0 des \u00e9conomies dans les soins de sant\u00e9 pour la population canadienne et ailleurs dans le monde, allant de l\u2019exactitude du premier diagnostic \u00e0 la r\u00e9duction des effets ind\u00e9sirables des m\u00e9dicaments. On reconna\u00eetra encore mieux le r\u00f4le de chef de file du Canada en analyse biologique.
