La compr\u00e9hension de l\u2019expression des g\u00e8nes est essentielle au progr\u00e8s de la m\u00e9decine : le niveau d\u2019expression de g\u00e8nes sp\u00e9cifiques peut indiquer si une personne peut entrer dans les premiers stades de la maladie. Quotidiennement, des chercheurs en g\u00e9nomique comprennent plus clairement les liens entre l\u2019expression des g\u00e8nes et la sant\u00e9 humaine.<\/p>\n<p>Les recherches sur les micro-ARN \u2013 une cat\u00e9gorie d\u2019acides nucl\u00e9iques d\u00e9couverte il y a \u00e0 peine plus de dix ans \u2013 ont permis de mieux comprendre la fa\u00e7on dont l\u2019expression des g\u00e8nes chez une personne refl\u00e8te son \u00e9tat de sant\u00e9. Les technologies puissantes utilis\u00e9es dans d\u2019autres domaines de la g\u00e9nomique pour quantifier le mat\u00e9riel g\u00e9n\u00e9tique se sont toutefois av\u00e9r\u00e9es lourdes, pour ne pas dire enti\u00e8rement inadapt\u00e9es \u2013 pour mesurer les micro-ARN. Ces difficult\u00e9s ont emp\u00each\u00e9 ce domaine de recherche de progresser encore plus rapidement. Par exemple, il peut falloir des dizaines d\u2019heures et co\u00fbter des dizaines de milliers de dollars pour mesurer une seule empreinte des niveaux d\u2019expression des micro-ARN.<\/p>\n<p>Nous proposons \u00e0 G\u00e9nome Canada un partenariat \u2013 en concertation avec la Fondation canadienne de recherche sur le cancer de la prostate, la Fondation de recherches de l\u2019Ontario, l\u2019Universit\u00e9 de Toronto, et la Canadian Microelectronics Corporation \u2013 pour b\u00e2tir une plateforme qui conviendrait id\u00e9alement \u00e0 la quantification des niveaux d\u2019expression des micro-ARN. Contrairement aux m\u00e9thodes existantes, la technologie que nous proposons de d\u00e9velopper co\u00fbtera moins de 10 $ par essai et n\u00e9cessitera une heure de temps d\u2019exploitation, et une autre heure de temps de mesure. Notre technologie offrira la sensibilit\u00e9 n\u00e9cessaire pour utiliser des \u00e9chantillons d\u00e9j\u00e0 disponibles dans les banques de tissus. Elle fournira la gamme dynamique n\u00e9cessaire \u00e0 la quantification des niveaux d\u2019expression fond\u00e9s sur les micro-ARN qui sont pertinents en g\u00e9nomique et en recherche clinique.<\/p>\n<p>Contrairement aux plateformes existantes \u2013 dans lesquelles interviennent le plus souvent diverses \u00e9tapes d\u2019amplification enzymatique et qui impliquent souvent la mesure d\u2019une lumi\u00e8re \u00e9mise par des sondes fluorescentes \u2013 notre puce sera purement \u00e9lectronique. Elle doit simplement mesurer le courant qui passe dans un \u00ab pixel \u00bb sp\u00e9cifique \u00e0 un micro-ARN, en r\u00e9ponse \u00e0 la pr\u00e9sence ou \u00e0 l\u2019absence de la paire compl\u00e9mentaire de ce brin, la cible recherch\u00e9e. Les puces de cam\u00e9ra \u00e0 5 $ qui se trouvent dans presque tous les t\u00e9l\u00e9phones cellulaires vendus aujourd\u2019hui peuvent d\u00e9tecter jusqu\u2019\u00e0 cinq \u00e9lectrons par pixel; nos circuits ont besoin d\u2019une sensibilit\u00e9 beaucoup moins grande, n\u2019exigeant que l\u2019\u00e9quivalent de 1 000 \u00e9lectrons d\u2019intensit\u00e9 de courant par pixel pour r\u00e9v\u00e9ler les donn\u00e9es congruentes en recherche. Les chercheurs du projet s\u2019efforceront de rendre leurs recherches pertinentes pour la communaut\u00e9 des plateformes de G\u00e9nome Canada par des appels t\u00e9l\u00e9phoniques trimestriels au Conseil consultatif de d\u00e9veloppement des technologies, compos\u00e9s d\u2019un biologiste-chimiste de renomm\u00e9e internationale chez Caltech; d\u2019un chef de file mondial canadien en g\u00e9nomique et en prot\u00e9omique et de deux experts de l\u2019industrie aux ant\u00e9c\u00e9dents exceptionnels parce qu\u2019ils ont su transformer des innovations de la recherche en des applications commerciales pertinentes en recherche clinique. Nous travaillerons en \u00e9troite collaboration avec la communaut\u00e9 de recherche clinique, notamment le Dr Fei-Fei Liu, oncologue clinicien \u00e0 l\u2019h\u00f4pital Princess Margaret.<\/p>\n<p>La chef de projet, Mme Shana Kelley et le cochercheur Ted Sargent veulent commercialiser la technologie. Le prototype \u00e9labor\u00e9 avec l\u2019aide de G\u00e9nome Canada consistera en un syst\u00e8me qui peut servir \u00e0 d\u00e9montrer la technologie pour obtenir le financement en capital de risque de s\u00e9rie A. Mme Kelley a d\u00e9j\u00e0 particip\u00e9 \u00e0 la recherche de financement d\u2019une entreprise prosp\u00e8re de diagnostics cliniques qui s\u2019est vendue 232 millions de dollars en 2006.<\/p>\n<p>M.
