Les progr\u00e8s r\u00e9cents des technologies de s\u00e9quen\u00e7age de l\u2019ADN changent la fa\u00e7on dont les chercheurs colligent et interpr\u00e8tent les donn\u00e9es en g\u00e9nomique. Au Genome Sciences Centre (GSC), nous int\u00e9grons les progr\u00e8s de la technologie et parvenons \u00e0 influencer favorablement le co\u00fbt, la qualit\u00e9 et la quantit\u00e9 des donn\u00e9es g\u00e9nomiques. Le Projet du g\u00e9nome humain, par exemple, a co\u00fbt\u00e9 trois milliards de dollars et il a fallu environ 15 ans pour le r\u00e9aliser. \u00c9tonnamment, en peu de temps seulement apr\u00e8s l\u2019ach\u00e8vement du projet de s\u00e9quen\u00e7age du g\u00e9nome humain, les progr\u00e8s technologiques r\u00e9cents visent \u00e0 rendre bient\u00f4t possible le \u00ab g\u00e9nome \u00e0 1000 dollars \u00bb.<\/p>\n<p>Les progr\u00e8s r\u00e9volutionnaires de cette technologie de pointe ne s\u2019appliquent pas seulement au s\u00e9quen\u00e7age de g\u00e9nomes entiers, mais procurent \u00e9galement une infrastructure indispensable aux chercheurs qui veulent comprendre des maladies g\u00e9n\u00e9tiques comme le cancer, le diab\u00e8te, les maladies cardiovasculaires et les maladies mentales, de m\u00eame que des probl\u00e8mes environnementaux actuels comme la biorestauration et les infestations d\u2019insectes ravageurs dans les for\u00eats.<\/p>\n<p>Gr\u00e2ce \u00e0 cette nouvelle technologie, nous pouvons pour la premi\u00e8re fois, par exemple, imaginer un deuxi\u00e8me s\u00e9quen\u00e7age des g\u00e9nomes humains et utiliser cette capacit\u00e9 pour comparer des r\u00e9gions du g\u00e9nome chez des individus en bonne sant\u00e9 et des individus malades afin de d\u00e9terminer les g\u00e8nes qui ont mut\u00e9 et qui causent la maladie. Nous pouvons \u00e9galement s\u00e9quencer des populations de microorganismes dans des \u00e9chantillons de sol ou d\u2019eau, ou encore les g\u00e9nomes d\u2019esp\u00e8ces importantes en agriculture. Ce type d\u2019information peut s\u2019appliquer \u00e0 un meilleur traitement des maladies, \u00e0 des strat\u00e9gies de pr\u00e9vention ou \u00e0 des th\u00e9rapies pharmaceutiques personnalis\u00e9es dans le cas des humains, et \u00e0 une meilleure lutte contre les insectes ravageurs en agriculture ou en foresterie, ou encore \u00e0 des m\u00e9thodes plus s\u00fbres pour le nettoyage de la contamination au p\u00e9trole, au moyen de microbes naturels, pour ne nommer que ces quelques applications.<\/p>\n<p>Cette technologie diff\u00e8re grandement des appareils et des m\u00e9thodes actuelles de s\u00e9quen\u00e7age et suppose l\u2019utilisation de nouveaux produits chimiques, la mise au point de m\u00e9thodes novatrices, la cr\u00e9ation de protocoles valid\u00e9s et le d\u00e9veloppement de nouvelles approches computationnelles pour traiter et interpr\u00e9ter les donn\u00e9es produites par ces nouveaux appareils. C\u2019est l\u00e0 un sujet d\u2019int\u00e9r\u00eat important du GSC depuis que nous avons fait l\u2019acquisition d\u2019un appareil de s\u00e9quen\u00e7age d\u2019acc\u00e8s rapide (appel\u00e9 Genome Analyzer) d\u2019Illumina en novembre 2006. Nous avons travaill\u00e9 consid\u00e9rablement avec Illumina pour produire une quantit\u00e9 substantielle de donn\u00e9es de s\u00e9quen\u00e7age de haute qualit\u00e9 (plus de 30 milliards de paires de base de s\u00e9quen\u00e7age), et nous avons \u00e9t\u00e9 extr\u00eamement impressionn\u00e9s par la quantit\u00e9 de donn\u00e9es de haute qualit\u00e9 qu\u2019il est possible de produire \u00e0 faible co\u00fbt et en assez peu de temps gr\u00e2ce \u00e0 cette technologie. Pour donner une id\u00e9e des \u00e9conomies de temps et d\u2019argent que permettraient de r\u00e9aliser les \u00ab s\u00e9quenceurs de la prochaine g\u00e9n\u00e9ration \u00bb, ces instruments peuvent produire plus d\u2019un milliard de paires de base de donn\u00e9es de s\u00e9quen\u00e7age tous les trois jours, alors que la g\u00e9n\u00e9ration actuelle d\u2019appareils ne produit qu\u2019environ trois millions de paires de base tous les trois jours.<\/p>\n<p>Cette diff\u00e9rence est \u00e9norme : un appareil de s\u00e9quen\u00e7age d\u2019Illumina peut d\u00e9terminer, en une seule ann\u00e9e, une quantit\u00e9 de s\u00e9quences d\u2019ADN qui \u00e9quivaut approximativement \u00e0 trois fois la quantit\u00e9 totale des s\u00e9quences produites dans le monde au moyen de centaines d\u2019appareils jusqu\u2019en janvier 2007. En plus de cette augmentation remarquable du taux de s\u00e9quen\u00e7age, les nouveaux appareils sont \u00e9galement 300 fois plus rentables.<\/p>\n<p>La pr\u00e9sente proposition d\u00e9crit un plan de deux ans qui misera sur la capacit\u00e9 de ces appareils de s\u00e9quen\u00e7age de la prochaine g\u00e9n\u00e9ration et d\u00e9veloppera des applications utiles pour un large spectre d\u2019\u00e9tudes biologiques et m\u00e9dicales.<\/p>\n<p>Nous serons en position id\u00e9ale pour r\u00e9aliser ces travaux au GSC en raison de notre expertise dans les domaines de la g\u00e9nomique, de la prot\u00e9omique et de la bioinformatique. Nous avons \u00e9tabli une installation de s\u00e9quen\u00e7age du g\u00e9nome \u00e0 haut d\u00e9bit dans le contexte de la plateforme de technologies Genome BC\/G\u00e9nome Canada, o\u00f9 nous appuyons des projets locaux, nationaux et internationaux de pointe. Pour demeurer concurrentiels \u00e0 l\u2019\u00e9chelle internationale, il est important pour nous de poursuivre ces efforts pionniers dans le d\u00e9veloppement des technologies et d\u2019\u00e9laborer de nouveaux protocoles et de nouveaux logiciels dans les domaines pr\u00e9cit\u00e9s.
