Notre laboratoire a r\u00e9cemment fait la preuve de principe d\u2019un prototype de \u00ab r\u00e9acteur prot\u00e9omique \u00bb qui permet de traiter avec efficacit\u00e9 d\u2019infimes quantit\u00e9s de prot\u00e9ines. Ce r\u00e9acteur simplifie la manipulation des prot\u00e9ines, r\u00e9duit consid\u00e9rablement les volumes n\u00e9cessaires \u00e0 l\u2019analyse et raccourcit la dur\u00e9e du traitement. Il a suscit\u00e9 beaucoup d&rsquo;int\u00e9r\u00eat aupr\u00e8s de soci\u00e9t\u00e9s canadiennes et internationales qui fabriquent de l&rsquo;\u00e9quipement scientifique.<\/p>\n<p>Les am\u00e9liorations dans le domaine de l\u2019analyse des prot\u00e9ines portent principalement sur l\u2019accroissement de la sensibilit\u00e9. Au cours des derni\u00e8res ann\u00e9es, les grandes am\u00e9liorations du rendement de techniques comme la spectrom\u00e9trie de masse et la fluorescence induite par laser ne se sont pas enti\u00e8rement traduites par une sensibilit\u00e9 accrue parce les \u00e9chantillons ne sont pas suffisamment trait\u00e9s avant l&rsquo;analyse. C&rsquo;est l\u00e0 l&rsquo;un des grands d\u00e9fis \u00e0 relever en prot\u00e9omique.<\/p>\n<p>Notre prototype de r\u00e9acteur repr\u00e9sente l\u2019appareil de traitement le plus efficace en prot\u00e9omique sur les plans de la sensibilit\u00e9, de la vitesse, et de la simplification du processus. Nous avons d\u00e9montr\u00e9 qu&rsquo;il est possible de traiter et d&rsquo;analyser par spectrom\u00e9trie de masse aussi peu que 300 cellules vivantes et que m\u00eame avec un nombre sup\u00e9rieur de cellules, le r\u00e9acteur offre un meilleur rendement que les m\u00e9thodes traditionnelles de traitement des prot\u00e9ines. Nous croyons que nous pouvons am\u00e9liorer ce rendement, d\u00e9velopper des r\u00e9acteurs qui donneront de meilleurs r\u00e9sultats prot\u00e9omiques pour les \u00e9chantillons prot\u00e9omiques courants et les \u00e9tudes des prot\u00e9ines jusqu&rsquo;au niveau des cellules individuelles. On aura ainsi de nouvelles applications comme la prot\u00e9omique des cellules individuelles, ce qui aura d&rsquo;\u00e9normes r\u00e9percussions en recherche sur les cellules souches et en m\u00e9decine r\u00e9g\u00e9n\u00e9rative.<\/p>\n<p>Nous proposons, dans ce projet, de poursuivre le d\u00e9veloppement et l\u2019expansion de la technologie du r\u00e9acteur prot\u00e9omique en deux phases. Dans une premi\u00e8re phase, nous augmenterons rapidement la taille actuelle du r\u00e9acteur prot\u00e9omique, en construisant quatre tailles de colonnes, de fa\u00e7on \u00e0 r\u00e9pondre \u00e0 des besoins d&rsquo;\u00e9chantillons qui vont de grands prot\u00e9omes complexes comme le plasma aux cellules individuelles. Des stations fluidiques automatis\u00e9es augmenteront la vitesse et l\u2019efficacit\u00e9 de ces appareils. D\u2019ici la deuxi\u00e8me ann\u00e9e, nous d\u00e9velopperons des ensembles de r\u00e9acteur prot\u00e9omique automatis\u00e9s et feront la preuve d&rsquo;applications novatrices en analyse cellule par cellule.<\/p>\n<p>Dans cette deuxi\u00e8me phase, nous transposerons et transformerons le r\u00e9acteur prot\u00e9omique \u00e0 l&rsquo;aide de la technologie de microfabrication du Conseil national de recherches. Ces puces microfluidiques am\u00e9lioreront encore davantage le traitement des \u00e9chantillons en r\u00e9duisant les volumes de r\u00e9action, en am\u00e9liorant l\u2019efficacit\u00e9 de la manipulation des \u00e9chantillons, de l&rsquo;automatisation et d\u2019une diff\u00e9rente biochimie de surface. Nous pr\u00e9voyons avoir termin\u00e9 la mise au point de trousses de r\u00e9acteur prot\u00e9omique \u00e0 puces avant la fin de la deuxi\u00e8me ann\u00e9e.<\/p>\n<p>Des s\u00e9ances de formation pour les chercheurs universitaires et les plateformes de G\u00e9nome Canada sont pr\u00e9vues au cours de la deuxi\u00e8me ann\u00e9e et dans l\u2019ann\u00e9e qui suivra la fin du projet.<\/p>\n<p>La mise au point de trousses de r\u00e9acteur prot\u00e9omique dot\u00e9s de colonnes de tailles diff\u00e9rentes et de r\u00e9acteur prot\u00e9omique \u00e0 puces am\u00e9liorera l\u2019efficacit\u00e9 de la recherche prot\u00e9omique dans les analyses qui vont du plasma aux cellules individuelles.<\/p>\n<p>Nous pr\u00e9voyons que le traitement de cellules individuelles dans le r\u00e9acteur, combin\u00e9e \u00e0 la d\u00e9tection par fluorescence, assurera une sensibilit\u00e9 qui nous permettra de mesurer les changements des profils prot\u00e9omiques dans des cellules individuelles.
