Cette s\u00e9quence a \u00e9t\u00e9 suivie d\u2019une v\u00e9ritable mar\u00e9e de s\u00e9quences bact\u00e9riennes et maintenant plus de 1 000 ont \u00e9t\u00e9 r\u00e9alis\u00e9es, ce qui \u00e9tablit la puissance du s\u00e9quen\u00e7age \u00e0 l\u2019aveugle appliqu\u00e9 avec succ\u00e8s \u00e0 de nombreux organismes, dont les humains, les singes, les chevaux, les chiens et d\u2019autres formes de vie.<\/p>\n<p>Parmi les s\u00e9quences bact\u00e9riennes, celle de <em>Rhodococcus<\/em> <em>jostii <\/em>RHA1, est remarquable, parce qu\u2019elle a \u00e9t\u00e9 la premi\u00e8re s\u00e9quence de g\u00e9nome r\u00e9alis\u00e9e au Canada et, jusqu\u2019\u00e0 r\u00e9cemment, le g\u00e9nome de la plus grosse bact\u00e9rie (9,7 Mb, &gt; 9 000 g\u00e8nes) \u00e9lucid\u00e9. Le projet a commenc\u00e9 en 2000 et a \u00e9t\u00e9 l\u2019\u0153uvre enti\u00e8re de l\u2019\u00e9quipe situ\u00e9e \u00e0 l\u2019Universit\u00e9 de la Colombie-Britannique qui s\u2019est charg\u00e9e du s\u00e9quen\u00e7age, de la bioinformatique et des analyses de g\u00e9nomique fonctionnelle telles que les \u00e9tudes du transcriptome et du prot\u00e9ome.<\/p>\n<p><em>R. jostii<\/em> RHA1 est digne de mention parce qu\u2019il s\u2019agit d\u2019un microbe environnemental d\u2019une grande utilit\u00e9. On l\u2019a qualifi\u00e9 de \u00ab centrale \u00bb de fonction de d\u00e9gradation parce qu\u2019il est capable de m\u00e9taboliser un tr\u00e8s large \u00e9ventail de mol\u00e9cules biologiques. Pendant le projet, notre \u00e9quipe et nos collaborateurs ont \u00e9tabli le fondement de la capacit\u00e9 de <em>R. jostii<\/em> RHA1 de d\u00e9grader une grande vari\u00e9t\u00e9 de polluants dont les biph\u00e9nyles polychlor\u00e9s (BPC), les nitriles, les phtalates, la dim\u00e9thylnitrosamine et les \u00e9thers diph\u00e9nyliques polybrom\u00e9s (EDP). La disponibilit\u00e9 de la s\u00e9quence compl\u00e8te a r\u00e9v\u00e9l\u00e9 le potentiel de <em>R. jostii <\/em>RHA1 dans de nombreuses applications, dont des applications commerciales pour le nettoyage de l\u2019environnement et les biotransformations de produits importants tels que les st\u00e9ro\u00efdes. La reconnaissance d\u2019une grappe de g\u00e8nes impliqu\u00e9s dans la d\u00e9gradation du cholest\u00e9rol, un type de st\u00e9ro\u00efde de grande distribution, a \u00e9t\u00e9 un avantage inattendu des \u00e9tudes en g\u00e9nomique de <em>R. jostii <\/em>RHA1. L\u2019analyse d\u00e9taill\u00e9e a effectivement r\u00e9v\u00e9l\u00e9 que cette grappe de g\u00e8nes \u00e9tait \u00e0 peu pr\u00e8s identique \u00e0 une autre grappe pr\u00e9sente dans le g\u00e9nome de la bact\u00e9rie <em>Mycobacterium<\/em> tuberculosis , agent pathog\u00e8ne mortel qui a tu\u00e9 plus d\u2019humains que tout autre organisme pathog\u00e8ne. L\u2019identification des fonctions de d\u00e9gradation du cholest\u00e9rol dans l\u2019agent responsable de la tuberculose a cr\u00e9\u00e9 la possibilit\u00e9 de nouvelles th\u00e9rapies pour cette terrible maladie.<\/p>\n<p><em>R. jostii<\/em> RHA1 peut servir dans d\u2019autres applications : il peut, par exemple, \u00eatre h\u00f4te de substitution pour la production d\u2019antibiotiques et l\u2019exploration de g\u00e8nes d\u2019organismes non cultiv\u00e9s dans divers environnements. RHA1 fait \u00e9galement l\u2019objet d\u2019autres \u00e9tudes approfondies \u00e0 mesure que l\u2019\u00e9quipe de l\u2019Universit\u00e9 de la Colombie- Britannique poursuit ses travaux sur ce microbe fascinant.
