Cette t\u00e2che est grandement compliqu\u00e9e par le fait que chez les mammif\u00e8res, un seul g\u00e8ne peut produire diverses isoformes prot\u00e9iques (formes mol\u00e9culaires multiples de prot\u00e9ines donn\u00e9es) par le biais d\u2019un processus appel\u00e9 \u00ab \u00e9pissage alternatif pr\u00e9- ARNm \u00bb. Les anomalies de l\u2019\u00e9pissage alternatif expliqueraient, croit-on, plusieurs types de cancer et d\u2019autres maladies humaines comme la fibrose kystique, la thallass\u00e9mie, et la maladie d&rsquo;Aran-Duchenne. Malgr\u00e9 sa pr\u00e9valence, on sait toutefois tr\u00e8s peu de choses sur les m\u00e9canismes biologiques qui r\u00e9gulent l\u2019\u00e9pissage alternatif.<\/p>\n<p>M. Sherif Abou-Elela, biologiste mol\u00e9culaire \u00e0 la facult\u00e9 de m\u00e9decine de l\u2019Universit\u00e9 de Sherbrooke est le directeur du projet Annotation fonctionnelle des isoformes essentielles alternativement \u00e9piss\u00e9es. En collaboration avec une \u00e9quipe de chercheurs hautement qualifi\u00e9s de la r\u00e9gion de Sherbrooke, M. Abou-Elela dirigera les travaux sur l\u2019annotation exp\u00e9rimentale des isoformes alternativement \u00e9piss\u00e9es dans quelque 600 g\u00e8nes li\u00e9s au cancer qui r\u00e9gulent la prolif\u00e9ration et la viabilit\u00e9 des cellules. L\u2019\u00e9quipe du projet \u00e9tudiera \u00e9galement les ph\u00e9notypes qui d\u00e9coulent de l\u2019inhibition sp\u00e9cifique aux isoformes, et mettra les donn\u00e9es obtenues en corr\u00e9lation avec des analyses d\u2019\u00e9chantillons de tissus provenant de populations canadiennes.<\/p>\n<p>Selon M. Abou-Elela, une autorit\u00e9 en g\u00e9nomique fonctionnelle \u00e0 haut rendement, l\u2019\u00e9quipe de Sherbrooke est le seul groupe au monde \u00e0 \u00e9tudier \u00e0 une si grande \u00e9chelle les annotations fonctionnelles des isoformes \u00e9piss\u00e9es li\u00e9es au cancer, de m\u00eame que les circuits de r\u00e9gulation qui les contr\u00f4lent. Ce projet d\u2019avant-garde \u00e0 l\u2019\u00e9chelle internationale recrutera, formera et maintiendra en poste un personnel hautement qualifi\u00e9. Les travaux sur l\u2019identification des marqueurs de l\u2019\u00e9pissage alternatif devraient en outre mener \u00e0 la mise au point de nouveaux outils de diagnostic utilis\u00e9s en clinique.
