Plus nous en savons sur les g\u00e8nes qui sont \u00e0 la base des maladies, plus nous pouvons intelligemment concevoir de nouveaux traitements. Il est rare qu\u2019une maladie g\u00e9n\u00e9tique soit simplement due \u00e0 une erreur dans un g\u00e8ne. Lorsque le d\u00e9faut d\u2019un seul g\u00e8ne peut \u00eatre la cause premi\u00e8re, la complexit\u00e9 du corps humain garantit presque que d\u2019autres g\u00e8nes seront en cause. Un exemple typique est le g\u00e8ne qui, lorsqu\u2019il fait d\u00e9faut, cause la fibrose kystique.<\/p>\n<p>Gr\u00e2ce aux travaux novateurs r\u00e9alis\u00e9s dans les ann\u00e9es 1980 par des chercheurs de l\u2019H\u00f4pital pour enfan ts malades, nous savons que le g\u00e8ne CFTR fait d\u00e9faut chez tous les patients atteints de fibrose kystique; ce g\u00e8ne commande \u00e0 la cellule de fabriquer une prot\u00e9ine qui d\u00e9place les ions chlore ordinaires \u00e0 l\u2019int\u00e9rieur et \u00e0 l\u2019ext\u00e9rieur des cellules. Pourtant, la gravit\u00e9 de la fibrose kystique peut \u00eatre tr\u00e8s diff\u00e9rente pour des patients chez qui la modification g\u00e9n\u00e9tique du g\u00e8ne CFTR est identique. L\u2019interaction d\u2019autres g\u00e8nes avec le g\u00e8ne CFTR doit expliquer cette modification du cours de la maladie.<\/p>\n<p>Nous avions pour objectif d\u2019utiliser les m\u00e9thodes les plus perfectionn\u00e9es de la g\u00e9nomique pour mettre au jour le plus grand nombre possible de ces autres g\u00e8nes. Ce type d\u2019\u00e9tude est le plus fructueux lorsqu\u2019il existe une \u00e9troite collaboration entre les m\u00e9decins et les chercheurs. Il faut des diagnostics et une description des sympt\u00f4mes de la maladie tr\u00e8s consciencieux. L\u2019\u00e9tude doit aussi pouvoir compter sur une solide organisation de base pour identifier les patients et leurs familles au Canada, obtenir d\u2019eux des \u00e9chantillons pour les analyses g\u00e9n\u00e9tiques, enregistrer et analyser ensuite les r\u00e9sultats.<\/p>\n<p>Nous avons collabor\u00e9 avec les 38 cliniques canadiennes de la fibrose kystique pour constituer un groupe d\u2019\u00e9tude repr\u00e9sentant plus de 75 % de toute la population canadienne atteinte de fibrose kystique. Ensemble, nous avons recueilli des pr\u00e9l\u00e8vements sanguins aupr\u00e8s de quelque 2 800 familles touch\u00e9es par la fibrose kystique, certaines de l\u2019ext\u00e9rieur du Canada. Nous avons extrait l\u2019ADN de ces pr\u00e9l\u00e8vements et avons cr\u00e9\u00e9 des \u00e9chantillons de cultures tissulaires qui peuvent \u00eatre conserv\u00e9s pour d\u2019autres \u00e9tudes; il s\u2019agit actuellement de la ressource mondiale la plus consid\u00e9rable pour les \u00e9tudes g\u00e9n\u00e9tiques sur la fibrose kystique. Notre analyse de l\u2019ADN extrait \u00e0 l\u2019aide de techniques g\u00e9n\u00e9tiques perfectionn\u00e9es a permis d\u2019identifier presque 100 g\u00e8nes susceptibles d\u2019influencer la progression de la fibrose kystique.<\/p>\n<p>&nbsp;<\/p>\n","protected":false},"excerpt":{"rendered":"<p>Un aspect important du Projet du g\u00e9nome humain r\u00e9side dans la promesse d\u2019une meilleure compr\u00e9hension et d\u2019un meilleur traitement des maladies humaines.
