Dans ce projet, les chercheurs se sont fix\u00e9s comme objectif d\u2019exploiter leurs connaissances de ces organismes mod\u00e8les pour comprendre la biologie des cellules humaines normales et malades. Dans la premi\u00e8re partie de leur \u00e9tude, les chercheurs ont caract\u00e9ris\u00e9 les g\u00e8nes et les prot\u00e9ines de la levure <em>Saccharomyces cerevisiae<\/em> . Ils documentent les effets des mutations dans chacun des g\u00e8nes sur les mol\u00e9cules (appel\u00e9es m\u00e9tabolites), qui fournissent aux cellules les signaux, l\u2019\u00e9nergie et les \u00e9l\u00e9ments const itutifs des structures cellulaires et consignent les effets des mutations sur la croissance, l\u2019apparence et la viabilit\u00e9 de l\u2019organisme.<\/p>\n<p>Gr\u00e2ce \u00e0 des technologies existantes et \u00e0 de nouvelles technologies que les chercheurs vont mettre au point, ces derniers purifient les prot\u00e9ines de la levure et d\u2019autres organismes, notamment la bact\u00e9rie<em> E.coli<\/em>. Ils savent cr\u00e9er des structures tridimensionnelles de haute r\u00e9solution des prot\u00e9ines, ils \u00e9tudient l\u2019interaction de ces prot\u00e9ines afin de d\u00e9couvrir la fonction des prot\u00e9ines. Pour brosser un tableau plus complet de la fonction de ces prot\u00e9ines, les chercheurs d\u00e9terminent \u00e9galement quels m\u00e9tabolites ou autres mol\u00e9cules se joignent \u00e0 chacune des prot\u00e9ines. En comprenant la biologie \u00e9l\u00e9mentaire d\u2019une cellule mod\u00e8le, nous apprendrons \u00e0 conna\u00eetre comme jamais auparavant les prot\u00e9ines de cellules humaines. Des scientifiques de l\u2019Universit\u00e9 de Toronto dirigent ce projet et travailleront en \u00e9troite collaboration avec plusieurs centres et consortiums aux \u00c9tats-Unis, de m\u00eame qu\u2019avec le RIKEN Genome Sciences Center, au Japon.<\/p>\n<p>UN R\u00c9SUM\u00c9 DES R\u00c9SULTATS DE CE PROJET TERMIN\u00c9 EST ACTUELLEMENT EN COURS DE PR\u00c9PARATION.
