Cependant, certains types de dommages de l\u2019ADN permettent que des cellules endommag\u00e9es continuent \u00e0 vivre et \u00e0 prolif\u00e9rer\u2014et ces cellules g\u00e9n\u00e9tiquement endommag\u00e9es finissent par causer le cancer, qui entra\u00eene une croissance cellulaire incontr\u00f4l\u00e9e et l\u2019invasion d\u2019autres sites du corps.<\/p>\n<p>La recherche sur le cancer est actuellement ax\u00e9e sur la d\u00e9tection des changements g\u00e9n\u00e9tiques li\u00e9s \u00e0 l\u2019apparition et la progression du cancer. De nouvelles technologies, mises au point \u00e0 la suite du projet du g\u00e9nome humain, permettent aux chercheurs d\u2019examiner des milliers de g\u00e8nes \u00e0 la fois pour d\u00e9terminer les alt\u00e9rations qui peuvent \u00eatre li\u00e9es au cancer. \u00ab Ces alt\u00e9rations g\u00e9n\u00e9tiques peuvent \u00eatre tr\u00e8s sp\u00e9cifiques dans le cas de certains types de cancer, explique le D<sup>r <\/sup>Douglas Horsman du B.C. Cancer Agency. Nous les utilisons pour faciliter le diagnostic du cancer, d\u00e9terminer si un cancer est b\u00e9nin ou malin, et comprendre si le malade r\u00e9agira \u00e0 certains types de th\u00e9rapie. Dans l\u2019avenir, l\u2019\u00e9volution de ces technologies m\u00e8nera \u00e0 la mise au point de th\u00e9rapies g\u00e9niques contre le cancer. \u00bb<\/p>\n<p>Dirig\u00e9 par les D<sup>rs<\/sup> Horsman et Wan Lam, le projet Mise au point et validation de micropuces d\u2019hybridation g\u00e9nomique comparative (CGH) en vue de leur utilisation clinique pour le cancer vise \u00e0 \u00e9laborer et \u00e0 \u00e9valuer une nouvelle technologie appel\u00e9e hybridation g\u00e9nomique comparative \u00e0 l\u2019aide de micropuces de tout le g\u00e9nome (aCGH). Cette technologie permet aux chercheurs sur le cancer d\u2019analyser tout l\u2019ADN de cellules canc\u00e9reuses afin d\u2019identifier des sites o\u00f9 il peut y avoir un changement de la quantit\u00e9 d\u2019ADN. Les r\u00e9gions de l\u2019ADN o\u00f9 l\u2019on constate un gain ou une perte anormale peuvent indiquer l\u2019emplacement de g\u00e8nes responsables de l\u2019apparition ou de la progression du cancer.<\/p>\n<p>Le projet comprendra une collaboration entre le BC Cancer Research Centre, le BC Cancer Agency et d\u2019autres centres de recherche sur le cancer d\u2019Edmonton et de Toronto. \u00ab Cette technique d\u00e9coule d\u2019un proc\u00e9d\u00e9 novateur bas\u00e9 sur le g\u00e9nome dans le cadre duquel on applique de petits fragments d\u2019ADN qui repr\u00e9sentent tout le g\u00e9nome humain sur des lamelles de microscope au moyen d\u2019une imprimante sp\u00e9ciale, puis des teintures qui \u00e9mettent une lumi\u00e8re color\u00e9e, afin d\u2019identifier les s\u00e9quences d\u2019ADN compl\u00e9mentaire provenant de sp\u00e9cimens canc\u00e9reux, explique le D<sup>r<\/sup> Horsman. \u00c0 l\u2019aide de cam\u00e9ras sp\u00e9ciales, l\u2019intensit\u00e9 de la lumi\u00e8re color\u00e9e \u00e9mise par chaque point sur la lamelle indique s\u2019il y a gain ou perte d\u2019ADN dans les cellules tumorales. \u00bb<\/p>\n<p>\u00c0 l\u2019aide de programmes informatiques, les chercheurs comparent ensuite les identit\u00e9s des fragments alt\u00e9r\u00e9s avec la s\u00e9quence du g\u00e9nome humain pour d\u00e9terminer quels g\u00e8nes se trouvent dans ces fragments.<\/p>\n<p>Jusqu\u2019\u00e0 maintenant, le prototype de la technologie CGH pang\u00e9nomique (appel\u00e9e micropuce SMRT) a \u00e9t\u00e9 d\u00e9velopp\u00e9 au BC Cancer Research Center et au Genome Sciences Centre, gr\u00e2ce au soutien permanent de G\u00e9nome Canada qui a investi dans le projet intitul\u00e9 \u00ab G\u00e9nomique du cancer : approche multidisciplinaire \u00e0 haut rendement visant l\u2019identification des g\u00e8nes impliqu\u00e9s dans les premiers stades du cancer \u00bb. Dans la poursuite des travaux, l\u2019\u00e9quipe du projet \u00e9laborera et validera maintenant la technologie aCGH aux fins de recherche et d\u2019application clinique courante. La technologie aCGH pang\u00e9nomique int\u00e9grera d\u2019autres techniques de recherche actuellement en cours d\u2019\u00e9laboration qui, en bout de ligne, transformeront l\u2019investigation et la gestion des patients atteints de cancer.
