Tous les processus de la vie commencent par la lecture de l\u2019information g\u00e9n\u00e9tique du g\u00e9nome (l\u2019ADN). La lecture des g\u00e8nes est magnifiquement complexe : elle se fait par le truchement d\u2019une classe de mol\u00e9cules appel\u00e9es ARN messagers, qui subissent de nombreuses modifications pendant qu\u2019elles remplissent leurs r\u00f4les de directeurs des activit\u00e9s de la cellule. Les anomalies dans ces modifications peuvent causer des maladies comme le cancer.<\/p>\n<p>La lecture de l\u2019information g\u00e9n\u00e9tique de presque chaque g\u00e8ne humain est soumise \u00e0 une modification particuli\u00e8re appel\u00e9e \u00e9pissage alternatif des ARN pr\u00e9messagers. Nous sommes des experts de l\u2019\u00e9pissage de l\u2019ARN et nous utilisons ces connaissances pour mieux comprendre comment les changements dans l\u2019expression des g\u00e8nes chez les humains peuvent mener \u00e0 des maladies. Nous connaissons \u00e9galement tr\u00e8s bien les processus chimiques de r\u00e9alisation d\u2019ARN en \u00e9prouvette pour cr\u00e9er de nouveaux moyens de lutter contre les infections bact\u00e9riennes.<\/p>\n<p>Nous avons mis au point une m\u00e9thode que nous appelons LISA (syst\u00e8me stratifi\u00e9 et int\u00e9gr\u00e9 d\u2019annotation de l\u2019\u00e9pissage). Cette m\u00e9thode nous permet d\u2019\u00e9tudier l\u2019\u00e9pissage alternatif de l\u2019ARN dans de nombreux g\u00e8nes en m\u00eame temps et de verser nos r\u00e9sultats dans une base de donn\u00e9es structur\u00e9e. Nous avons apport\u00e9 des modifications chimiques \u00e0 l\u2019ARN qui nous permettent d\u2019\u00e9tudier l\u2019\u00e9pissage alternatif; globalement, \u00e0 l\u2019aide de LISA, nous avons d\u00e9crit l\u2019\u00e9pissage alternatif dans presque 100 g\u00e8nes importants dans la maladie du cancer. La m\u00e9thode LISA et la base de donn\u00e9es sont \u00e0 la disposition d\u2019autres chercheurs partout dans le monde. Nous avons fabriqu\u00e9 des d\u00e9riv\u00e9s chimiques de mol\u00e9cules d\u2019ARN qui nous permettent d\u2019entrer facilement dans les cellules, en particulier les bact\u00e9ries.
