On ne dispose pas cependant d\u2019outils g\u00e9n\u00e9tiques pour un grand nombre d\u2019organismes et m\u00eame les plus simples manipulations g\u00e9nomiques demeurent difficiles. Le clonage de chromosomes entiers ou de grands fragments d\u2019ADN du g\u00e9nome du donneur dans des souches h\u00f4tes substituts comme la levure, o\u00f9 des manipulations g\u00e9n\u00e9tiques sont possibles, peut att\u00e9nuer consid\u00e9rablement ces limites.<\/p>\n<p>MM.\u00a0Turlough Finan, Bogumil Karas et Trevor Charles, Ph.\u00a0D., mettent au point un syst\u00e8me d\u2019h\u00f4tes bact\u00e9riens substituts (<em>Sinorhizobium meliloti<\/em>) qui permet de reproduire et de cr\u00e9er de larges fragments d\u2019ADN et de les transf\u00e9rer efficacement dans des cellules h\u00f4tes bact\u00e9riennes et eucaryotes, un transfert qui n\u2019est actuellement pas possible avec la levure. Ainsi, de grands fragments d\u2019ADN ou des chromosomes entiers seront clon\u00e9s dans <em>S.\u00a0meliloti<\/em>, cr\u00e9\u00e9s et r\u00e9introduits dans l\u2019organisme d\u2019origine. En plus de son application g\u00e9n\u00e9rale en ing\u00e9nierie des g\u00e9nomes, la technologie de l\u2019h\u00f4te substitut <em>S.\u00a0meliloti<\/em> peut \u00eatre utilis\u00e9e pour mettre au point \u00e0 br\u00e8ve \u00e9ch\u00e9ance des technologies, notamment la production de grandes biblioth\u00e8ques d\u2019ADN m\u00e9tag\u00e9nomiques pour la bioprospection.<\/p>\n<p>Le projet fera acqu\u00e9rir une expertise en biologie synth\u00e9tique au Canada, ax\u00e9e sur la manipulation du g\u00e9nome complet et l\u2019utilisation d\u2019un syst\u00e8me h\u00f4te substitut en ing\u00e9nierie des g\u00e9nomes.
