En raison de leur r\u00f4le important dans la mise au point des m\u00e9dicaments et la recherche biom\u00e9dicale, le nombre et la diversit\u00e9 de mod\u00e8les murins continuent d\u2019augmenter \u00e0 un rythme sans pr\u00e9c\u00e9dent. Toutefois, de nouveaux mod\u00e8les murins exigent une validation approfondie pour d\u00e9terminer s\u2019ils repr\u00e9sentent v\u00e9ritablement la maladie humaine et une caract\u00e9risation compl\u00e8te pour acqu\u00e9rir de nouvelles connaissances sur la sant\u00e9 et la maladie, proposer des approches de traitement et faciliter la mise au point de m\u00e9dicaments. La d\u00e9termination de la concentration d\u2019une grande vari\u00e9t\u00e9 de prot\u00e9ines dans les fluides et les tissus biologiques des souris est un moyen important d\u2019\u00e9valuer ces mod\u00e8les, mais les m\u00e9thodes actuelles ne peuvent \u00e9tudier \u00e0 la fois que de petits nombres des quelque 16\u00a0000\u00a0prot\u00e9ines que contient une souris. Une approche plus exhaustive est indispensable pour faire progresser ce domaine.<\/p>\n<p>M. Christoph Borchers, Ph.\u00a0D., et son \u00e9quipe de l\u2019Universit\u00e9 de Victoria, en collaboration avec leur partenaire, M.\u00a0Albert Sickmann, Ph.\u00a0D., en Allemagne (ISAS), effectueront les premi\u00e8res \u00e9tapes cruciales de la quantification panprot\u00e9omique dans les souris. L\u2019\u00e9quipe du projet mettra au point une nouvelle technologie bas\u00e9e sur la spectrom\u00e9trie de masse pour l\u2019analyse rapide de 3\u00a0000\u00a0prot\u00e9ines murines, soit environ 20\u00a0% du nombre total de prot\u00e9ines, dans 20\u00a0tissus diff\u00e9rents. La technologie sera fournie \u00e0 titre de service et format\u00e9e dans une trousse facile d\u2019utilisation \u00e0 l\u2019intention des chercheurs de partout dans le monde. L\u2019\u00e9quipe publiera \u00e9galement des fourchettes de r\u00e9f\u00e9rence qui aideront les chercheurs \u00e0 interpr\u00e9ter les donn\u00e9es obtenues \u00e0 l\u2019aide des trousses. Cette technologie \u00e9largira la port\u00e9e scientifique et permettra d\u2019obtenir un tableau plus approfondi et complet du r\u00f4le jou\u00e9 par l\u2019ensemble des prot\u00e9ines contenues dans le g\u00e9nome, que le sujet soit en sant\u00e9 ou malade.
