Ce m\u00e9canisme d\u00e9pend d\u2019une petite prot\u00e9ine appel\u00e9e ubiquitine, qui se lie aux prot\u00e9ines cibles et signale leur destruction. Ce processus est indispensable au fonctionnement normal des cellules. Toutefois, pour de nombreuses maladies, l\u2019ubiquitine n\u2019est pas li\u00e9e ou d\u00e9truite comme elle le devrait, ce qui entra\u00eene une d\u00e9gradation anormale des prot\u00e9ines. La fixation ou la destruction appropri\u00e9es d\u00e9pendent de certaines enzymes qui ont r\u00e9cemment retenu l\u2019attention dans la mise au point de nouveaux m\u00e9dicaments parce qu\u2019on a observ\u00e9 qu\u2019elles fonctionnaient mal dans plusieurs maladies. Quelques m\u00e9dicaments actuellement approuv\u00e9s exercent un effet sur ces enzymes et ont \u00e9t\u00e9 utilis\u00e9s avec succ\u00e8s dans le traitement de certains types de cancer, mais ils ont des effets ind\u00e9sirables. Globalement, il y a peu de mol\u00e9cules qui influencent la fonction de ces enzymes, ce qui a limit\u00e9 les tentatives qui pouvaient \u00eatre faites pour les manipuler \u00e0 des fins th\u00e9rapeutiques.<\/p>\n<p>Sachdev Sidhu, Ph.\u00a0D., de l\u2019Universit\u00e9 de Toronto dirige une \u00e9quipe qui utilisera une plateforme \u00e0 haut rendement de g\u00e9nie g\u00e9n\u00e9tique et mol\u00e9culaire novatrice, unique dans le monde. Elle a d\u2019ailleurs suscit\u00e9 beaucoup d\u2019int\u00e9r\u00eat dans l\u2019industrie et les milieux universitaires parce qu\u2019elle permet la mise au point rapide et \u00e9conomique de mol\u00e9cules tr\u00e8s sp\u00e9cifiques et puissantes, semblables \u00e0 l\u2019ubiquitine, pour se fixer \u00e0 des enzymes cl\u00e9s li\u00e9es au cancer, de fa\u00e7on \u00e0 bloquer ou \u00e0 am\u00e9liorer leur fonction. L\u2019effet biologique de ces mol\u00e9cules sera valid\u00e9 dans des mod\u00e8les cellulaires et animaux du cancer. Le projet produira des mol\u00e9cules pr\u00e9cliniques pr\u00eates pour la mise au point de produits th\u00e9rapeutiques.
