Pour effectuer leurs fonctions, les prot\u00e9ines doivent s\u2019associer les unes aux autres par des interactions prot\u00e9ine-prot\u00e9ine ou IPP. Les prot\u00e9ines membranaires int\u00e9grales sont un type important de prot\u00e9ines qui joue un r\u00f4le dans de nombreuses maladies humaines, mais depuis toujours difficiles \u00e0 \u00e9tudier en raison de leurs caract\u00e9ristiques biochimiques exceptionnelles. Ces caract\u00e9ristiques rendent extr\u00eamement difficile la connaissance de leurs interactions fonctionnelles avec d\u2019autres prot\u00e9ines et de ce qui fait en sorte que la perturbation de ces fonctions m\u00e8ne \u00e0 la maladie, ce qui repr\u00e9sente un obstacle important pour les chercheurs qui veulent cr\u00e9er de nouveaux m\u00e9dicaments pour la traiter.<\/p>\n<p>Igor Stagljar, Ph. D., \u00a0et son \u00e9quipe de l\u2019Universit\u00e9 de Toronto, ont r\u00e9cemment mis au point une nouvelle technologie puissante appel\u00e9e Test double hybride de membranes de mammif\u00e8res (MaMTH), qui peut cartographier les IPP de prot\u00e9ines membranaires int\u00e9grales directement dans le contexte naturel d\u2019une cellule. Cette perc\u00e9e constitue une avanc\u00e9e importante dans l\u2019\u00e9tude des prot\u00e9ines membranaires chez les humains. L\u2019\u00e9quipe propose maintenant de perfectionner la technologie MaMTH en la transformant en une plateforme qui peut cartographier les interactions de prot\u00e9ines membranaires int\u00e9grales \u00e0 une \u00e9chelle extr\u00eamement grande, permettant ainsi d\u2019identifier et d\u2019\u00e9tudier les IPP de membranes int\u00e9grales \u00e0 un rythme sans pr\u00e9c\u00e9dent. Ces travaux fourniront une nouvelle technologie g\u00e9nomique transformatrice, de m\u00eame qu\u2019un grand volume d\u2019information sur les interactions des prot\u00e9ines membranaires int\u00e9grales gr\u00e2ce auquel les chercheurs pourront mieux comprendre la fonction des prot\u00e9ines membranaires et mettre au point plus rapidement des th\u00e9rapies mieux cibl\u00e9es pour les maladies humaines.<\/p>\n<p>De plus, cette technologie sera le fondement d\u2019une entreprise ontarienne appel\u00e9e Protein Network Sciences qui aidera les chercheurs \u00e0 progresser en leur facilitant l\u2019acc\u00e8s \u00e0 cette nouvelle technologie de rupture MaMTH.
