L\u2019interaction des trois types de cellules peut dicter l\u2019\u00e9volution du cancer au fil du temps, de m\u00eame que la r\u00e9action ou la r\u00e9sistance aux traitements. Des progr\u00e8s r\u00e9cents des technologies de s\u00e9quen\u00e7age microfluidique et de s\u00e9quen\u00e7age de l\u2019ADN ont permis aux chercheurs d\u2019analyser simultan\u00e9ment des dizaines de milliers de cellules individuelles provenant de tissus complexes, y compris des tumeurs. L\u2019interpr\u00e9tation de ces donn\u00e9es est difficile en raison du manque d\u2019ensembles de r\u00e9f\u00e9rences de haute qualit\u00e9 pour chaque type de cellule dans le corps et l\u2019absence de m\u00e9thodes pour relier ces donn\u00e9es \u00e0 la biologie des tumeurs.<\/p>\n<p>Trevor Pugh, Ph. D., du Princess Margaret Cancer Centre et Michael Brudno, Ph. D., de l\u2019Hospital for Sick Children mettent au point la trousse d\u2019outils d\u2019expression des cellules du cancer \u00e0 l\u2019\u00e9chelle de la cellule unique (CReSCENT), un ensemble \u00e9volutif et normalis\u00e9 de m\u00e9thodes algorithmiques, d\u2019outils et de portail de donn\u00e9es nouveaux mis au point dans une infrastructure nuagique. Pour comparer les cellules dans des tissus canc\u00e9reux et sains, le syst\u00e8me regroupera les donn\u00e9es g\u00e9nomiques d\u2019une cellule unique, produites par des chercheurs sp\u00e9cialistes du cancer, et les reliera \u00e0 des donn\u00e9es de r\u00e9f\u00e9rence internationales produites par des experts de partout dans le monde dans le cadre de l\u2019atlas des cellules humaines. Ce syst\u00e8me de partage et de regroupement des donn\u00e9es est un facteur cl\u00e9 de diff\u00e9renciation de CReSCENT qui augmentera la productivit\u00e9 des chercheurs en acc\u00e9l\u00e9rant l\u2019ex\u00e9cution et la comparaison des m\u00e9thodes informatiques, tout en fournissant des donn\u00e9es contextuelles qui faciliteront la compr\u00e9hension du comportement des cellules dans des tissus tumoraux.<\/p>\n<p>Cette plateforme, que tout chercheur sur n\u2019importe quelle plateforme informatique pourra utiliser, constituera une ressource de donn\u00e9es cruciale pour la prochaine vague de recherches en g\u00e9nomique du cancer \u00e0 l\u2019\u00e9chelle de la cellule unique.
