Il arrive que l\u2019environnement ou l\u2019exposition modifie le fonctionnement des g\u00e8nes. La m\u00e9thylation de l\u2019ADN est une des causes de modification de l\u2019activit\u00e9 g\u00e9n\u00e9tique. Il s\u2019agit d\u2019un processus lors duquel une mol\u00e9cule de m\u00e9thyle se lie \u00e0 un nucl\u00e9otide cytosine dans l\u2019ADN. Des m\u00e9canismes pr\u00e9cis de m\u00e9thylation sont n\u00e9cessaires au d\u00e9veloppement normal; chez l\u2019humain, la perturbation de la m\u00e9thylation joue un r\u00f4le dans certaines maladies, comme le cancer et les maladies auto-immunes. Depuis peu, il est techniquement et financi\u00e8rement possible de mesurer la m\u00e9thylation de l\u2019ADN avec une r\u00e9solution de la taille d\u2019un nucl\u00e9otide simple, \u00e0 une vaste \u00e9chelle dans tout le g\u00e9nome, au moyen d\u2019une m\u00e9thode qu\u2019on appelle \u00ab s\u00e9quen\u00e7age au bisulfite \u00bb. Toutefois, les donn\u00e9es obtenues par ce s\u00e9quen\u00e7age pr\u00e9sentent de nombreuses mesures impr\u00e9cises ou manquantes, ce qui les rend difficiles \u00e0 interpr\u00e9ter et limite le potentiel des \u00e9tudes pour d\u00e9crire le r\u00f4le de l\u2019\u00e9pig\u00e9n\u00e9tique dans une maladie.<\/p>\n<p>Celia Greenwood, Ph. D., de l\u2019Institut Lady Davis de recherches m\u00e9dicales, et Karim Oualkacha, Ph. D., de l\u2019Universit\u00e9 du Qu\u00e9bec \u00e0 Montr\u00e9al, ont r\u00e9uni une \u00e9quipe d\u2019experts en vue de cr\u00e9er un ensemble comprenant un algorithme et un logiciel pour analyser des donn\u00e9es sur la m\u00e9thylation de l\u2019ADN \u00e0 grande \u00e9chelle et de grande dimension, afin que nous puissions profiter du potentiel cach\u00e9 de ces donn\u00e9es. Les m\u00e9thodes et la trousse se pencheront d\u2019abord sur la scl\u00e9rodermie, une maladie auto-immune invalidante causant la cicatrisation de nombreux tissus et dont la r\u00e9ponse aux traitements est impr\u00e9visible.<\/p>\n<p>L\u2019\u00e9quipe a le soutien d\u2019un organisme de d\u00e9fense des patients, Scl\u00e9rodermie Qu\u00e9bec, et de deux entreprises de Montr\u00e9al, une ayant mis au point des m\u00e9thodes d\u2019apprentissage machine et l\u2019autre fournissant une plateforme informatique \u00e0 haut rendement.
