L\u2019administration de vaccins vivants peut permettre de ma\u00eetriser la coccidiose, mais la mise en \u0153uvre d\u2019une telle mesure peut s\u2019av\u00e9rer difficile. Ce type de vaccination repose sur l\u2019utilisation de parasites infectieux vivants pour causer une infection subclinique spontan\u00e9ment r\u00e9solutive qui stimule le d\u00e9veloppement d\u2019une immunit\u00e9 protectrice robuste. L\u2019exactitude de la dose est essentielle \u00e0 la r\u00e9ussite de la vaccination; l\u2019administration d\u2019un trop grand nombre de parasites infectieux aura un impact n\u00e9gatif sur la sant\u00e9 des oiseaux, tandis que l\u2019administration d\u2019un trop petit nombre ne stimulera pas une immunit\u00e9 protectrice. L\u2019efficacit\u00e9 des vaccins n\u2019est pas statique et d\u00e9pend de nombreuses variables. Chaque lot de vaccins doit \u00eatre mis \u00e0 l\u2019essai sur des animaux vivants afin de confirmer l\u2019infectiosit\u00e9 et la dose correcte. Les essais sur les infections prennent beaucoup de temps, co\u00fbtent cher et ne sont, au mieux, que semi-quantitatifs. Nous avons \u00e9valu\u00e9 une \u00e9preuve prototype qui permet une \u00e9valuation rapide de la viabilit\u00e9 des parasites (c.-\u00e0-d. de l\u2019efficacit\u00e9 du vaccin) fond\u00e9e sur la mesure de l\u2019activit\u00e9 mol\u00e9culaire r\u00e9elle de ces parasites. Nous avons mis au point un protocole permettant de mesurer les biomol\u00e9cules produites par les parasites pr\u00e9sents dans les vaccins lors de l\u2019exposition \u00e0 des stimuli particuliers et nous avons montr\u00e9 que l\u2019abondance de ces cibles particuli\u00e8res refl\u00e8te la viabilit\u00e9 r\u00e9elle des parasites. Les travaux en cours visent \u00e0 am\u00e9liorer la pr\u00e9cision de l\u2019\u00e9preuve en d\u00e9terminant des cibles biomol\u00e9culaires optimis\u00e9es et \u00e0 rationaliser l\u2019\u00e9preuve pour en faciliter l\u2019utilisation. D\u2019autres travaux appuyant la mise au point d\u2019\u00e9preuves am\u00e9lioreront encore l\u2019accessibilit\u00e9 et la faisabilit\u00e9 de la vaccination en permettant le d\u00e9veloppement d\u2019outils mol\u00e9culaires normalis\u00e9s pour l\u2019identification rapide des esp\u00e8ces et souches parasitaires (utiles pour cerner les raisons de l\u2019\u00e9chec de la vaccination) et en prolongeant la dur\u00e9e de conservation des vaccins entrepos\u00e9s. Notre \u00e9preuve de viabilit\u00e9 optimis\u00e9e permettra de mesurer pr\u00e9cis\u00e9ment la viabilit\u00e9 des agents contenus dans les vaccins vivants contre la coccidiose, et ce, en quelques heures (au lieu des 10\u00a0jours ou plus requis dans les essais standards sur les infections). Cette d\u00e9couverte, combin\u00e9e aux outils mol\u00e9culaires susmentionn\u00e9s que nous d\u00e9veloppons \u00e0 l\u2019appui de l\u2019\u00e9preuve, am\u00e9liorera grandement l\u2019efficacit\u00e9 et l\u2019accessibilit\u00e9 des vaccins contre la coccidiose. Cela maximisera l\u2019efficacit\u00e9 de la production, les b\u00e9n\u00e9fices des agriculteurs et la durabilit\u00e9 de cette industrie alimentaire d\u2019importance mondiale, tout en r\u00e9duisant au minimum l\u2019impact sur l\u2019environnement.
